Opioid peptides are peptides that bind to opioid receptors in the brain; opiates and opioids mimic the effect of these peptides. Such peptides may be produced by the body itself, for example endorphins. The effects of these peptides vary, but they all resemble those of opiates. Brain opioid peptide systems are known to play an important role in motivation, emotion, attachment behaviour, the response to stress and pain, and the control of food intake.

The endogenous peptides with opiate-like activity. The three major classes currently recognized are the ENKEPHALINS, the DYNORPHINS, and the ENDORPHINS. Opioid peptides are endogenous substances that exhibit the pharmacological properties of morphine. Morphine (named after Morpheous, the Greek god of dreams) is an alkaloid analgesic, isolated as one of the components of opium, the infamous product of the poppy, Papaver somniferum. The opioid peptides are involved in physiological actions such as motor behavior, feeding, drinking, analgesia, immune responses, memory, and in the regulation of gastrointestinal, cardiovascular, neuronendocrine, and cognitive functions.

Opioid peptides produced in the body act as neuromodulators that modify the actions of other neurotransmitters in the central nervous system. By altering the electrical properties of their target neurons, thereby making these neurons more difficult to excite, opioid peptides can influence the release of various neurotransmitters. As a result of this modulation, opioid peptides can-among other functions-induce pain relief and euphoria as well as affect certain behaviors, including alcohol consumption. Alcohol can activate the opioid peptide system.

Twenty-six papers by leading researchers survey opioid peptides and their receptors and discuss in detail how such receptors regulate both transmitter and hormone release as well as integrative mechanisms in the central nervous system. The contributions emphasize their modulation of neural excitability and their control of cardiovascular mechanisms. They also illustrate how receptor-type selective opioids may have therapeutic implications other than the classic ones.
